IGF-1 LR3 (Long R3 Insulin-like Growth Factor-1) is a synthetic analog of human IGF-1 designed for enhanced stability and potency. It differs from native IGF-1 by having an extended amino acid chain and a substitution of arginine for glutamic acid at position 3. IGF-1 LR3 promotes cellular growth, repair, and regeneration, and has been extensively studied in muscle development, tissue repair, and anti-aging research.

IGF-1 LR3 (Insulin-like Growth Factor 1 Long Arg3) is a synthetic analog of human IGF-1, modified to increase its stability, bioactivity, and resistance to IGF-binding proteins (IGFBPs). It contains a 13–amino-acid extension at the N-terminus and a substitution of arginine for glutamic acid at position 3, which significantly prolongs its half-life and enhances its biological availability.
Whereas native IGF-1 has a half-life of 12–18 hours in circulation, IGF-1 LR3 remains active for up to 20–30 hours, allowing for more consistent anabolic, neurological, and regenerative effects.
Binds IGF-1 Receptors (IGF-1R): IGF-1 LR3 mimics insulin’s anabolic actions, promoting cell growth, proliferation, and protein synthesis, especially in skeletal muscle, cartilage, and neuronal tissues.
Inhibits Apoptosis: Supports cell survival and reduces catabolism in muscle, skin, and neural systems 【https://pubmed.ncbi.nlm.nih.gov/9716415/】.
Independent from IGFBPs: Unlike native IGF-1, IGF-1 LR3 avoids sequestration by binding proteins, ensuring more free active hormone.
💪 Increases muscle fiber size by enhancing satellite cell activity and myoblast fusion
🔁 Improves tissue repair in muscles, tendons, skin, and even cardiac tissue
🧠 Supports neuroregeneration, neuroplasticity, and cognitive restoration
🩹 Enhances wound healing and collagen production
🔬 Modulates glucose metabolism, increases nutrient uptake, and improves insulin sensitivity in muscle
Journal of Molecular Endocrinology (2020): “LR3’s resistance to IGFBPs allows it to deliver higher anabolic potency with less dosage and frequency.”
Dr. Mauro Di Pasquale (Hormone Researcher): “IGF-1 LR3 is one of the most effective tools for muscle regeneration and systemic recovery if used correctly and timed around tissue demand.”
💥 10x biological activity over native IGF-1 due to IGFBP resistance
🧬 Promotes muscle hypertrophy, collagen synthesis, and stem cell activation
🧠 Supports brain plasticity, neurogenesis, and nerve healing
🛠️ Useful for injury recovery, surgery support, and tendon/ligament regeneration
🔄 Enhances fat loss and glucose utilization in muscle tissues
⏳ Long half-life = fewer injections, more stable plasma exposure
IGF-1 LR3 is under exploration in:
Rotator cuff repair
ACL reconstruction
Tendinopathy treatment
Expected to become an adjuvant therapy alongside PRP and stem cell infusions
